Means 'now that ~; since ~ (has been decided/become the case)'. Because a certain situation or decision is now a fact, a natural conclusion, resolve, or obligation follows. It carries a firm, determined tone — the speaker commits to seeing things through given the established circumstances.

引き受けた上は、最後までやり遂げます。
Hikiuketa ue wa, saigo made yaritogemasu.
Now that I've taken this on, I'll see it through to the end.
引き受けた (accepted) + 上は; because the task was accepted, firm resolve follows.
留学すると決めた上は、しっかり準備しよう。
Ryuugaku suru to kimeta ue wa, shikkari junbi shiyou.
Now that I've decided to study abroad, let me prepare thoroughly.
決めた (decided) + 上は leads to a determined course of action.
約束した上は、必ず守るべきだ。
Yakusoku shita ue wa, kanarazu mamoru beki da.
Since you've made a promise, you should absolutely keep it.
約束した (promised) + 上は produces an obligation expressed with べき.
• The second clause expresses resolve, obligation, or a natural must (よう, べき, なければ, つもり). • Register: formal / written / decisive speech. • Nearly interchangeable with ~以上は and ~からには.
The JLPT rarely asks whether you recognise a pattern — it asks which of several similar patterns fits. These are the ones ~上は is tested against.
からには is very close and slightly more common in speech; ~上は is more formal and written, but both mean 'now that / since'.
上(で) marks a step done before another action ('after ~'); 上は marks an established fact from which resolve or obligation follows ('now that ~').
